I am not able to manually lock my doors on my 2025 Ram 150 Limited when I exit. If the lock button is pressed on either front door, the locks engage, but when the door is closed, the horn chirps 3 times and all the doors unlock. Truck doors lock fine with key FOB. Is that a some new safety feature? Can it be defeated?

I was able to manually lock my 2021, 2019 and 2016 Rams..

That to me sounds like you have a key still in the truck. Do you have both of your keys on you?

Key fobs are not the truck

Could be something with the programing. Have you thought of bring it in to the dealer and seeing if there is something they can do. Its under warranty right?

the 2025 rams have a super aggressive "key lockout protection" feature that prevents the doors from staying locked if the system detects the key fob might still be inside. when you hit the manual lock button and close the door, the truck pings for the fob; if it senses the signal is too close to the interior, it chirps three times and unlocks everything to save you from a lockout.

you can usually find the setting to tweak this in your uconnect 5 screen under vehicle > settings > doors & locks. look for "flash lights with lock" or "passive entry" settings, but most importantly, check if there is a "key-in-vehicle" alert you can toggle. since you have a limited with all the tech, the sensors are extra sensitive, so even having the fob in your pocket right next to the door as it closes can trigger it.
